- Smart-gel trapped particles were successfully developed as a delivery platform for delivering a combination of hydrophilic and hydrophobic drugs in combination management therapy. The purpose of this is to serve as an alternative for treatment of chronic conditions such as type 2 diabetes, where dosage forms often fail to meet expectations. Coenzyme Q10 loaded poly (lactide-co-glycolide) nanoparticles/microparticles (10mg) and insulin solution (3 mg/ml) were added into Chitosan (1% w/v) solution and vortexed for 5 min. Chilled β-glycerol phosphate (GP) (5.6%) solution was then added to the above solutions and was subsequently vortexed for 5 min. The resulting formulation containing IS and Co-Q10-PLGA-NPorMP was characterized for gelling time, content uniformity, rheology, texture analysis and TGA. These formulations were lyophilised and were characterised with TGA, DSC, XRD and SEM. In situ smart gelling system containing anti-oxidant hydrophobic CoQ10 loaded PLGA and hydrophilic IS was prepared successfully. Rheological characterisation showed these formulations as shear thinning systems/ pseudo plastic and is hence thixotropic. Texture analysis showed possession of a lower viscosity at the sol phase of the formulations in comparison to the gel phase. This allows ease of administration during the sol phase into the subcutaneous tissue; the system then behaves as an implant following gelling in vivo .DSC of this formulation (lyophilized powder) showed that Co-Q10 was in an amorphous form as compared with native Co-Q10, this result was supported by XRD analysis of the same formulation. SEM images showed the uniformly distributed NP in the C-GP powder in comparism with blank gel where nanoparticles are conspicuously absent. This study clearly demonstrates that the smart in situ gelling system containing anti-oxidant hydrophobic CoQ10-PLGA-NP and anti-diabetic hydrophilic IS can be used for the management of chronic diseases such as diabetes.
